Microarray, SAGE and their applications to cardiovascular diseases

    摘要 ThewealthofDNAdatageneratedbythehumangenomeprojectcouplingwithrecentlyinventedhigh-throughputgeneexpressionprofilingtechniqueshasdramaticallyspeduptheprocessforbiomedicalresearchersonelucidatingtheroleofgenesinhumandiseases.Onepowerfulmethodtorevealinsightintogenefunctionsisthesystematicanalysisofgeneexpression.Twopopularhigh-throughputgeneexpressiontechnologies,microarrayandSerialAnalysisofGeneExpression(SAGE)arecapableofproducinglargeamountsofgeneexpressiondatawiththepotentialofprovidingnovelinsightsintofundamentaldiseaseprocesses,especiallycomplexsyndromessuchascardiovasculardisease,whoseetiologiesareduetomultiplegeneticfactorsandtheirinterplaywiththeenvironment.MicroarrayandSAGEhavealreadybeenusedtoexaminegeneexpressionpatternsofcell-culture,animalandhumantissuesmodelsofcardiovasculardiseases.Inthisreview,wewillfirstgiveabriefintroductionofmicroarrayandSAGEtechnologiesandpointouttheirlimitations.Wewillthendiscussthemajordiscoveriesandthenewbiologicalinsightsthathaveemergedfromtheirapplicationstocardiovasculardiseases.Finallywewilltouchuponpotentialchallengesandfuturedevelopmentsinthisarea.
