Effect of Alstonia Scholaris bark extract on testicular function of Wistar rats

    摘要 Aim:ToevaluatetheantifertilityeffectofAlstoniascholarisbarkextractinmalerats.Methods:InmaleWistarratsAlstoniascholarisbarkextractwasgivenbyoralrouteatadoseof200mg/dayfor60days,Thefertilityandtesticularfunctionwereassessedbymatingtests,spermmotility,spermconcentration,biochemicalindicesandtesticularcellpopulationdynamics.Results:Oralfeedingwiththeextractatadoseof200mg/dayfortheperiodof60daysdidnotcausebodyweightloss,whiletheweightsoftestes,epididymides,seminalvesicleandventralprostateweresignificantlyreduced.Theproductionofstep-19spermatidswasreducedby79.6%intreatedrats.Thepopu-lationofpreleptoteneandpachytenespermatocytesweredecrasedby61.9%and60.1%,respectively.Spematogo-niaandSertolicellpopulationwerealsoaffected.TheseminiferoustubuleandLeydigcellnuclearareawerereducedsignificantly(P<0.01)whencomparedtothecontrols..Reducedspermcountandmotilityyesultedinatotalsupprs-sionoffertility.Asignificantfallintheproteinandsialicacidcontentofthetestes,epididymides,seminalvesicleandventralprostateaswellasglycogencontentoftesteswerealsonoticed.Thefructosecontentintheseminalvesiclewasloweredwhereasthetesticularcholesterolwaselevatedascomparedwiththecontrols.Thefollowingcom-poundswereobtainedfromtheextractwithchromatographicseparationoverSi-gelcolumn;α-amyrin,β-amyrin,lupiolacetate,venenative,rhazineandyohimbine,Conclusion:Atthedoselevelemployed,Alstoniascholarisbarkextracthasasignificantantifertilityeffectinmalerast;theprimarysiteofactionmaybepostmeioticgermcells(Step19spermatids).
